I would love it. I had Wario's Woods on SNES and I already love the fact that Mario Bros. 2 elements are included. I am all for old games getting represented. ^_^
It would be a nice nod, maybe they could appear in last levels, serving as the main antagonist's personal minions ? (the other ennemies are either influenced like the bosses or naturaly antagonistic to you)

At least some nods, Captain Toad is the first game starring a Toad since Wario's woods, i'd be disapointed if they don't make a reference to it.

Kirby is the best at surprise creepy. I would love to see this game start to get creepy at the end with maybe an optional boss that is like a nightmare.
I was thinking it's like a bit like the first king boo battle, the boss itself is creepy then when he get at low health the Bowser's head get in the wrong way and he go crazy...or like when Zero get at low health, he gorily rip his eye out...
Here the final boss is creepy but when he get at low health his mask break and he reveal his true face with his eyes starting to bleed.

Well SMB2 has quite a lot of nightmarish elements, since this game clearly take a bit of inspiration from it, i can see that they have a lot of creepy things which they could draw from it (Phanto, Hawkmouth, Wart bleeding from the eyes, maybe the masks from Doki doki panic...)
Also there was blood in the japanese version of PMTTYD...
While the villain could sport an incan inspired bird mask in a ghadius way, i would love to see his face being inspired by the third mask on the bottom-right (it even has red markings under the eyes which could be replaced by blood)
I notice also that Wingo's eyeslash are pretty similar to the one around Hawkmouth's eyes and the second mask in the pic above, so such antagonist would probably have indeed SMB2 carnival elements (i would love him to wear black lipstick, since the 3rd mask clearly show that black lipstick and white skin with markings would make him very cool looking)
Metroid could get dark with dead scientists and some bosses like Nightmare's face, or Mother Brain, as mentioned.
PM tend to have dark things, even SS had things like the rather cruel method of killings and a Toad who was in Black market.
Captain Toad should deal with things that are out of his world, he wander and find other unknown lands which have problems since Mario & co aren't even aware that they exist at all.
That Toad in the alley who rips you off in Sticker Star is also shady!
Some unknown lands should be better than the Mushroom Kingdom and like paradise while others should be dark and like a nightmare world. Both kinds should appear.
That would great too! A place could be like an illusion when observed from a distance to make the observer see what ever they view as paradise only to be heck when they land.
